Those holes are in the inner wheel-well. That can be fixed by fiberglassing over the holes, but you need some kind of backing behind the holes. You can use plastic sheets, or metal, to create something behind the holes for the fiberglass to hold to.
I ain't much on bodywerk, but I'm sure there is someone hear that can explain how to fix this issue.

From your original description, I think we were all thinking you had holes in the actual fender(s), which would still be repairable, but more involved.

My 1st thought when I saw the post was to shovel some dirt in it. Then I read where the dark side was creepin in on these dudes with the red Vettes....

Since it is your inner fender well you have a couple of options. Youcan remove it and replace it without alot of effort and maybe less time. To repair it you can take it out and lay glass over the backside and undercoat the front side back in.

Budget version is that you can loosen up the access bolts and slip a patch of plastic or sheet metal and pop rivet it in place and then undercoat the front side.

Just depends on hard you want to work, and how professional you want to be.

It's in a good hiding place and we won't tell but that NCRS judge won't be to forgiving for anything less than an OEM well....

Joe, since your goin to do the frame off put it on your replacement list that you will start. Give it a priority and price out a new one. Then as you start to rebuild you can go as far as the money goes. I'm sure you will be dealing with some more expensive stuff on your list. But if your goin thru all the resto might as well plan to do it right. Sounds like this vette wil be in the family for ever. Dad would be proud!Wink

You can reach up through the hole and clean up the back side around the hole.  A piece of fiberglass matting can be soaked with resin, and unfolded on the back side of the hole.  A piece of wire will help.  You may need to leave a small hole to reach the wire through and work it around to get the matting to lay aginst the back of the panel.   Then hold wax paper up on the front side to keep the mat from sagging toward you.  Give it a few minutes to set up and you now have a backing to work from.  
 
If you get to the back side without reaching through, it is easier, but still lay a layer of resin soaked matting on the inside.
 
Now from the outside you can put a couple more layers of glass in to complete the fill.  Sand down any extra sticking out, and finish off with a bit of filler.  Glass reinforced would be stronger.  Sand the surface smooth and paint or undercoat.
 
If you can reach the back, you may want to put two more layers of matting on the back side for extra strength, but if not okay.  Do this after the first layer sets.  That will hold it in place while it dries.    Good as new.
